This Technical Research Communication describes a new method by which thermally treated Coxiella in milk products may be grown in a liquid growth medium and quantitated using an MPN-PCR assay. Coxiella is generally not used in studies on thermal and non-thermal processing of milk due to the need for specialized and highly laborious techniques such as animal assays and tissue culture for determining viability. Recently, a liquid growth medium (ACCM-2) and modified atmosphere were used to grow Coxiella from pure cultures, infected mouse tissues, and clinical samples, however, the ability to grow Coxiella from a food such as milk has not been shown. The potential ability to enrich Coxiella directly from contaminated milk presents a new avenue for conducting pasteurization research in which the viability of heat-treated or injured cultures could be more easily determined through direct enrichment of Coxiella in ACCM-2. ACCM-2 medium allowed enrichment of Coxiella from bovine whole milk and cream, whole goat, and whole camel milks but not whole water buffalo milk. Enrichment was possible from whole bovine milk containing as few as 6 Coxiella ge/ml of milk. The applicability of this ACCM-2 enrichment method was shown when using an MPN-PCR assay to quantitate the number of viable Coxiella remaining in whole bovine milk after 64 °C thermal treatment for up to 10 min.
Microfaunal remains from a modern east African raptor roost: patterning and implications for fossil bone scatters

Fish eagles (genus Haliaeetus) potentially have contributed the bones of their prey to many late Cenozoic concentrations of vertebrate microfossils. To evaluate possible biases in the taxa and skeletal elements preserved in assemblages accumulated by fish eagles, which could affect paleoecological and evolutionary interpretations derived from Cenozoic microfaunal localities, a sample of 1883 bones accumulated by modern African fish eagles (H. vocifer) was collected from a roost near Lake Turkana, Kenya. Characteristics of the bone assemblage include (1) taxonomic composition dominated by fish; (2) diverse but small-sized and ecologically restricted animals including inshore fish, aquatic birds, very small nocturnal mammals (which may have been contributed by other raptors), and small reptiles (possible predators on the roost); (3) poor survivorship for specific categories of skeletal elements; (4) considerable bone breakage for all but the smallest animals; and (5) a distinctive pattern of postcranial fragmentation for bird and mammal elements. The fish eagle element profile has some similarities to those of other diurnal raptors, but its cumulative signature can be distinguished from those of other bone accumulators and includes characteristics that would also be manifest in fossil assemblages. Examination of fossil microfaunas from the Plio-Pleistocene localities of Kanapoi (Kenya), Inolelo (Tanzania), and Olduvai Gorge (Tanzania) shows that the patterns of element preservation for Kanapoi and Inolelo do not match those of the Lake Turkana fish eagle roost. Nonetheless, some aspects of the Olduvai Gorge FLKN assemblage show taxonomic and skeletal similarites. In light of the selective choices of prey by fish eagles, the fauna recovered from Olduvai Gorge FLKN might not accurately reflect faunal diversity or patterns of abundance at this locality during the Pleistocene. Because fish eagles of the genus Haliaeetus are found on three continents, have a fossil record extending to the Miocene, and deposit bones in a habitat conducive to preservation, the taxa preserved at many other late Cenozoic microfaunal sites also might be biased. Therefore, taphonomic profiles of late Cenozoic microfaunal assemblages should be compared against the taphonomic profiles typical of fish eagle roosts before faunal characterizations of sites are applied in analyses of paleoecological evolution.
Quantitation of viable Coxiella burnetii in milk using an integrated cell culture-polymerase chain reaction (ICC-PCR) assay

The obligate intracellular pathogen Coxiella burnetii has long been considered the most heat resistant pathogen in raw milk, making it the reference pathogen for determining pasteurisation conditions for milk products. New milk formulations and novel non-thermal processes require validation of effectiveness which requires a more practical method for analysis than using the currently used animal model for assessing Coxiella survival. Also, there is an interest in better characterising thermal inactivation of Coxiella in various milk formulations. To avoid the use of the guinea pig model for evaluating Coxiella survival, an Integrated Cell Culture-PCR (ICC-PCR) method was developed for determining Coxiella viability in milk. Vero cell cultures were directly infected from Coxiella-contaminated milk in duplicate 24-well plates. Viability of the Coxiella in milk was shown by a ≥0·5 log genome equivalent (ge)/ml increase in the quantity of IS111a gene from the baseline post-infection (day 0) level after 9–11 d propagation. Coxiella in skim, 2%, and whole milk, and half and half successfully infected Vero cells and increased in number by at least 2 logs using a 48-h infection period followed by 9-d propagation time. As few as 125 Coxiella ge/ml in whole milk was shown to infect and propagate at least 2 logs in the optimised ICC-PCR assay, though variable confirmation of propagation was shown for as low as 25 Coxiella ge/ml. Applicability of the ICC-PCR method was further proven in an MPN format to quantitate the number of viable Coxiella remaining in whole milk after 60 °C thermal treatment at 0, 20, 40, 60 and 90 min.

Identifying Barriers to Entering Smoking Cessation Treatment Among Socioeconomically Disadvantaged Smokers

Background: Efficacious smoking cessation interventions exist, yet few smokers utilise available resources such as psychosocial treatment programs and pharmacotherapy. The goals of the present study were to (1) identify perceived barriers to entering smoking cessation treatment programs among socioeconomically disadvantaged smokers, who are presently underrepresented in smoking cessation interventions; (2) determine what variables are most important in predicting the barriers identified (i.e., age, gender, ethnicity, income, nicotine dependence level, smoking rate, years smoking, stage of change, presence of smoking-related illness and medical insurance status). Methods: Responses from socioeconomically disadvantaged smokers (N = 343) were collected in 2004–2005 and analysed to develop the Treatment Barriers Questionnaire, a 40-item measure of reasons for not entering smoking cessation programs. Study methods were approved by the Institutional Review Board of Louisiana State University; informed consent procedures were employed. Results: Principal components analysis yielded seven scales named for their theme: (1) Preparedness to Quit Smoking; (2) Work and Time Constraints; (3) Smokers Can or Should Quit on Own; (4) Opinions about Professional Assistance; (5) Mobility Limitations; (6) Insurance Limitations and (7) Misinformation about Professional Assistance. Gender, ethnicity, daily smoking rate, nicotine dependence and stage of change were significant predictors in regression analyses for scales 1, F(10, 201) = 7.83, p < .001, R2 = .29, 2 F(10, 201) = 2.30, p < .05, R2 = .11, and 3, F(10, 201) = 3.58, p < .001, R2 = .16. Conclusions: Results can inform efforts to facilitate entry and retention of smokers in cessation programs.

Leg length, skull circumference, and the prevalence of dementia in low and middle income countries: a 10/66 population-based cross sectional survey

Background: Adult leg length is influenced by nutrition in the first few years of life. Adult head circumference is an indicator of brain growth. There is a limited literature linking short legs and small skulls to an increased risk for cognitive impairment and dementia in late life.
Methods: One phase cross-sectional surveys were carried out of all residents aged over 65 years in 11 catchment areas in China, India, Cuba, Dominican Republic, Venezuela, Mexico and Peru (n = 14,960). The cross-culturally validated 10/66 dementia diagnosis, and a sociodemographic and risk factor questionnaire were administered to all participants, and anthropometric measures taken. Poisson regression was used to calculate prevalence ratios for the effect of leg length and skull circumference upon 10/66 dementia, controlling for age, gender, education and family history of dementia.
Results: The pooled meta-analyzed fixed effect for leg length (highest vs. lowest quarter) was 0.82 (95% CI, 0.68–0.98) and for skull circumference 0.75 (95% CI, 0.63–0.89). While point estimates varied between sites, the proportion of the variability attributable to heterogeneity between studies as opposed to sampling error (I2) was 0% for leg length and 22% for skull circumference. The effects were independent and not mediated by family history of dementia. The effect of skull circumference was not modified by educational level or gender, and the effect of leg length was not modified by gender.
Conclusions: Since leg length and skull circumference are said to remain stable throughout adulthood into old age, reverse causality is an unlikely explanation for the findings. Early life nutritional programming, as well as neurodevelopment may protect against neurodegeneration.

Introduction: Cessation efforts and maintained abstinence in smokers have been associated with social support from others (i.e., ‘support persons’). Characteristics of support persons appear to affect the amount and quality of support provided to adults who smoke. In the present investigation, the relationship between support person characteristics and perceived quality and quantity of smoking-specific support provided was examined. Method: College students (N = 244) were asked to identify an adult who smokes about whom they were concerned. Participants reported demographic and smoking-related information about themselves and their identified smoker and perceptions of positive and negative smoking-specific social support provided. Results: Results indicated significant differences in smoking status (p < .05), such that current and ex-smokers reported providing more negative support than never smokers. Additionally, participants romantically involved with their smokers reported providing more positive (p < .05) support than those in other relationships. Discussion: Findings suggest the importance of relationship in the perception of support provided during the cessation process and highlight the need for future research in this area.
